Title: Redrilling a Ball Questions?

I may be buying a used ball on here that has already been plugged once. How would a second plug and redrilling effect ball reaction?

999 times out of 1000, hardly at all.
I've seen 3rd and 4th drills having no problems getting an excellent reaction from a ball. Of course that does not mean you won't be case #1000!

If the plug work is mostly in the same place, and the pro shop guy doing the work.  If he is good at it, you have no worries.
